Spectroscopic Studies on the Interaction of Co(II) Tetrapyridinoporphyrazine with Synthetic Polynucleotides and DNA

Interactions of cationic tetrakis (N,N´,N´´,N´´´-tetramethyltetra-3,4-pyridinoporphyrazinatocobalt(II) (Co(tmtppa)) with synthetic polynucleotides, poly(A-T), poly(G-C) and calf thymus DNA have been characterized in 5 mM phosphate buffer, pH 7.2, by optical absorption and fluorescence spectroscopy. The appearance of hypochromicity effect and the red shift in UV-Vis spectrum of porphyrazine was ...

A Study on the Aggregation and Calf Thymus DNA Binding Characteristics of Anionic Cobalt(II) Tetrasulfonated Phthalocyanine

The aggregation behavior of anionic Cobalt(II) 4,4′,4ʺ,4‴-tetrasulfonated phthalocyanine, [Co(TSPc)4-] was studied at its various concentrations and different ionic strengths using optical absorption and resonance light scattering (RLS) spectroscopies in 5 mM phosphate buffer, pH 7.0 at 25 °C. The results show no aggregation behavior at concentration range of 5.1 × 10-6-7....

Evaluation and Characterization of Free and Immobilized Acethylcholinesterase with Fluorescent Probe, Differential Scanning Calorimetry and Docking

Acetylcholinesterase (AChE) enzyme which catalyses the hydrolysis of choline esters, such as acetylcholine, is very important in nerve function. Previous structural studies showed the possible amyloid fibril formation on the AChE. Therefore it is important to understand interaction of ligands to prevent the formation of  amyloid fibrils. The purpose of the present study was to  char...

Comparative studies of the binding of ethidium bromide and its photoreactive analogues to nucleic acids by fluorescence and rapid kinetics.

The binding studies of the interaction of ethidium bromide with DNA have been hampered by its reversibility, which prevents direct isolation and thus characterization of the complex. The recent development of photoaffinity labeling has provided a means to circumvent this problem. However, to be useful as a probe for the parent compound, a photosensitive analogue must be shown to interact in viv...

of the extent of binding. This effect (known informally as "Chip dip") is illustrated in Figs. 6 and 7 which concern the ethidium bromide/tRNA system. 41 In this case the free ethidium bromide has a lifetime of 1.86 nsec and a rotational relaxation time of 0.54 nsec. On binding the PhetRNA Phe the lifetime increases to 26 nsec and the rotational relaxation increases to 136 nsec. A single rotati...
